Method

 

Material

• Heating agitator
• EDTA powder 95 %
• Toluidine blue 5 %
• Distilled water

 

Technic

1. Acid preparation :

· Mix 5 g of EDTA powder and 100 g of water
· Heat up to 200 °C with agitation
· Maintain heat and agitation until complete dissolution
· This weak acid looses its properties after one month

2. Apply a drop of 5 % EDTA on otolith for 3 minutes.

3. Rinse with distilled water.

4. Apply a drop of 5 % toluidine blue on otolith.

5. Leave the stain dry overnight.

6. Rinse with water.
